In situ detection and cloning of sequence specific DNA binding proteins in S. purpuratus sea urchin. Micro injection of fusion gene constructs into sea urchin eggs. DNA cloning, sequencing, PCR, recombineering, invitro transcription, translation, gel shifts, Northern, Southern and Western blots. Research Technician, Dept. of Basic and Clinical Research Scripps Clinic and Research Foundation. 1980 – 1984 Cloning of retroviral vector system for enzyme replacement therapy, retroviral expression in embryonal carcinoma cells, cloning and characterization of beta-glucosecerebrosidase gene (Gaucher's disease), protein purification and enzymatic assay. Molecular biological techniques, reverse transcriptase assay, tissue culture, fractionation of whole blood, hemoglobin and hematocrit determination.
